Quinidine - Induced Thrombocytopenia and Leukopenia : Demonstration and Characterization of Distinct Antiplatelet
نویسندگان
چکیده
A patient with the rare syndrome of simultaneous quinidine-induced thrombocytopenia and leukopenia was studied. A quinidine-dependent antiplatelet antibody was detected in her serum by platelet aggregometry and by indirect platelet-suspension immunofluorescence. A drugdependent antileukocyte antibody was demonstrated by leukoagglutination and by granulocyte immunofluorescence. Both antibodies were found to belong to the lgG class by immunofluorescence using monospecific antisera. There appeared to be two distinct antibodies. since the antibody eluted off sensitized platelets reacted only with platelets and that eluted off sensitized granulocytes

The mechanism of drug-dependent immunologic thrombocytopenic purpura (DITP) was investigated by studying the sera of four patients with classic DITP (two with quinidine-, one with acetaminophen-, and one with phenazopyridine-dependent antiplatelet antibody) using a solid-phase radioimmunoassay with 125I-staphylococcal protein A. Two forms of antiplatelet antibody could be demonstrated: one that...

The molecular nature of platelet receptors for quinineand quinidine-dependent antiplatelet antibodies (Q.Ab and Qd.Ab) was studied by immunoblotting. One Q.Ab caused quinine-dependent lgG binding to platelet proteins with molecular weights (mol wts) of 1 74 Kd and 93 Kd and another to only a 93-Kd protein. A third Q.Ab caused binding to 1 74-. 140-. 93-. and 57-Kd proteins, while a fourth Q.Ab ...

متن کاملQuinine - and Quinidine - dependent Antiplatelet Antibodies REQUIREMENT OF FACTOR VIII - RELATED ANTIGEN FOR PLATELET DAMAGE AND FOR IN VITRO TRANSFORMATION OF LYMPHOCYTES FROM PATIENTS WITH DRUG - INDUCED THROMBOCYTOPENIA SHARRON
PRP by drug-dependent antibodies were significantly reduced in comparison with PRP from normal controls. Addition ofpurified VIIIR:Ag restored levels ofplatelet damage to that of normal PRP. In vWd-PRP, platelet damage by two human antiplatelet sera, not dependent on drugs, and by a rabbit antiplatelet serum did not differ from that in normal PRP. PRP from patients deficient in Factor VIII coag...
